Nucifera C18H源于硅氧烷在硅胶表面进行单体自组装形成聚合物单分子层。引入十八烷基后,该固定相不仅具有出色的反相保留特性,还表现出良好的耐水能力,大幅改善了通用型C18固定相在高水相流动相中存在 “相坍塌(phase collapsing)”或“脱浸润(de-wetting)”现象。该固定相的栅栏结构可以保持十八烷基链在纯水中的舒展状态,富含硅醚的复合相结构抑制了硅羟基活性,为碱性物质的分离提供了便利。除了能够胜任通用型C18的分离分析任务,该固定相还适用于生物分子及其衍生物的高效分离,例如寡糖、氨基酸、短肽、核苷酸及有机酸。

Nucifera C18E色谱固定相表面键合有大位阻基团保护的十八烷基链,可以长期稳定地在pH 1.0-10.0的范围内工作。在确保大位阻复合基团有效保护十八烷基的情况下,汉德科技采用专利工艺使该固定相拥有与常规C18相似的碳含量,疏水保留能力不因键合量改变而改变。其他厂商提供的大位阻C18的碳含量通常为15%,Nucifera C18E为18%。该键合相的另一个特点是能够兼容任意水含量的反相流动相,包括100%水。当分析物在C18E上保留过强、分离度足够大时,可以更换为C8E以获得更高的分离速度。

Nucifera C18K色谱固定相的官能团为双齿键合的十八烷基硅氧烷,它与常规C18M和前述C18E反相特征一致,碳含量相仿,约为18%,具有优异的疏水性,适于疏水作用主导的反相色谱分离分析任务;双齿键合形式不仅提升了烷基链的有序性,使该填料具有较C18M和C18E更高的立体选择性(菲vs蒽),还极大地提高了固定相的耐碱能力,使其能够长期稳定地在pH 2.0-11.0的范围内工作,如此便可使用强碱性流动相以期改善某些碱性化合物,如阿米替林和普萘洛尔等叔胺类物质,在缓冲盐体系中因硅羟基次级氢键作用导致的保留过强、拖尾、峰形不对称的问题。低聚合度的硅氧烷键合相还能够兼容100%水,此时强极性化合物的保留得以显著增强,为分析人员提供更高的分离度和柱容量。当分析物在C18K上保留过强、分离度足够大时,可以更换为C8K以获得更高的分离速度。
