C4色谱柱,也称为丁基键合硅胶色谱柱,键合基团致密,疏水性良好,适合于分析亲脂性及双亲性化合物。C4在pH值2-8条件下相对稳定。它适于分子量超过1000的化合物,是蛋白和多肽的分离首选,当需要减少疏水性相互作用以避免非特异性吸附或蛋白质的聚集时,可以考虑使用C4色谱柱。
C8色谱柱的疏水保留能力较C4更强,能够分离C4上分离度较低的化合物。稳定性高于C4,能够在pH值2-9范围内使用。C8可广泛应用于疏水性有机化合物的分离和分析,如农药、多环芳烃、脂肪酸等,对极性较小的化合物具有较好的选择性,也适用于分离中等极性的化合物。此外,C8色谱柱与C18色谱柱相比,具有更低的疏水性,适合分离在C18柱上具有强吸附作用的化合物。在C18上保留过强的情况下,C8色谱柱通常表现出更高的分离效率及更短的分析时间。
